I'm sure that there will be away around this, if we can't summount these problems then how on earth are we even going to get anywhere in space? It is possible the timeline is a little optimistic, but if we don't try we will never suceed

i think the biggest problem to overcome would be the radiation...
i've read about creating artificial gravity. what they think may work would be only having a section of the ship rotate in a circle to create the gravitational effect, so they would spend most of their time in sections without gravity, whenever they workout they would enter this one section
i'm sure the food would not be too much of a problem
fueling would be another problem
